Neutralization of human IL-17A bioactivity. It is recommended to establish the optimal antibody concentration for the assay system used. The antibody is also suitable for immunoassays. Neutralization Neutralization assay: Neutralization of human IL-17A induced IL-6 production (see graph). To determine the neutralizing capacity and ND50, various concentrations of mAb MT504 and human recombinant IL-17A were added to normal human dermal fibroblasts. Following overnight incubation, cell supernatants were quantified in IL-6 ELISA. Neutralization dose 50 (ND50): MT504 neutralized the effect of IL-17A at a ND50value of 0.01-0.03 μg/mL when 10 ng/mL IL-17A was present. ND50 is defined as the concentration of antibody that results in 50 % inhibition of the biological response under the experimental conditions specified.
