Immunogen
- anti-Synaptotagmin 1 (SYT1) is an alpaca single-domain antibody (sdAb) that recognizes specifically the cytoplasmic domain of murine Synaptotagmin 1 with high affinity and specificity.
-
Labeling Ratio
- Two site-specifically conjugated fluorophores per sdAb.

-
Application Notes
- Optimal working dilution should be determined by the investigator.
-
Comment
-
Two site-specifically coupled fluorophores per molecule. The reagent can therefore simultaneously target two fluorophores to your protein of interest, which results in enhanced image brightness. Owing to the small size of the sdAb, the distance between the target epitope and each fluorophore is below 4 nm. In comparison to conventional detection systems using conventional antibodies, this sdAb can thus improve the localization accuracy by 10-15 nm. Both features - enhanced brightness and precise fluorophore placement - renders this product superior tools for all microscopy techniques.

Reconstitution
- Reconstitute with 50 % glycerol in deionized water. We recommend including 0.1 % sodium azide as a preservative if applicable.
-
Buffer
- lyophilized from PBS pH7.4 with 2% BSA (US-Origin)
-
Storage

Storage Comment
- Vials containing lyophilized protein can be stored at 4 °C for 6 months. We recommend reconstituting the protein with 50 % sterile glycerol including 0.1 % sodium azide as preservative if applicable. Minimize the number of freeze-thaw cycles by aliquoting the reconstituted protein. Long term storage at -80 °C for up to 6 months. Working aliquots can be stored at -20 °C for up to 4 weeks. We do not recommend storing the reconstituted protein at 4 °C.

Background
- Synaptotagmin 1 (Syt1) belongs to a family with 17 members. Synaptotagmins are all single-pass transmembrane proteins containing two domains binding to calcium at their intracellular portion. Synaptotagmin 1 is enriched at the pre-synaptic boutons on axon terminals, associated with synaptic vesicles, and is implicated in triggering vesicle fusion and neurotransmitter release upon a local increase of calcium concentration.
