Background
- Background: The insulin receptor substrate 1 (IRS1) is one of four members of the IRS family (IRS1-4) that mediates intracellular signaling downstream of the IGF1 and insulin receptors. Phosphorylation of IRS1 by activated receptors leads to binding of IRS to the SH2 domains of effector proteins such as PI3K, Grb2, nck, and SHP2. The downstream effects of insulin and IGF1 signaling via IRSs on the cell are pleiotropic and include various responses such as glucose transport, glycogen synthesis, protein synthesis, antilipolysis, and gluconeogenesis. Polymorphisms in IRS1 may contribute to the etiology of non-insulin-dependent diabetes mellitus (NIDDM).
