- Application Notes
- The original mouse antibody was able to bind fibrin even in the presence of human fibrinogen at the concentration found in plasma. This antibody when intravenously administered in a chicken ex vivo circulatory model bound human fibrin-coated disks placed in an extracorporeal chamber (PMID: 6648524). An immunoconjugate (tPA-59D8) generated from tissue plasminogen activator (tPA) and anti-fibrin antibody 59D8 degraded less fibrinogen and consumed less alpha 2-antiplasmin than did tPA alone, suggesting that more potent and more selective thrombolysis can be achieved by efficiently directing tPA to the site of a thrombus (PMID: 3118374). This antibody was used in the generation of a single-chain urokinase plasminogen activator (scu-PA) and -59D8 Fab' conjugate which had a fibrinolytic potency 33-fold more than that of tissue plasminogen activator (PMID: 2111744). A recombinant r-scuPA(32 kDa)-59D8 was 6 times more potent than high molecular mass scuPA in lysing a human plasma clot in vitro and was 20 times more potent than high molecular mass scuPA in the rabbit jugular vein model of thrombolysis. The binding affinity of the antibody towards fibrin was determined using ELISA (PMID: 1946453). This antibody was also used in the generation of a fusion protein comprising hirudin that genetically fused to the C-terminus of the variable region of the light chain of scFv59D8. This fusion protein was found to inhibit thrombin after its activation by factor Xa and also inhibits appositional clot growth in vitro (PMID: 10715263).
