    Specificity

    This antibody binds human IL-8, which is a chemotactic factor that attracts neutrophils, basophils, and T-cells, but not monocytes. It is also involved in neutrophil activation. It is released from several cell types in response to an inflammatory stimulus. This antibody cross reacts with rabbit IL-8.

    The original antibody was generated by immunizing BALB/c mice with human IL-8. This reshaped WS-4 antibody was generated by grafting CDRs of mouse monoclonal antibody onto human framework regions.

  • Application Notes

    This antibody was used in the generation of a very sensitive ELISA for the detection of human IL-8 in various body fluids. The original mouse version of this antibody has a detection limit of 16 pg/mL in an ELISA. This antibody can bind both the 72 and 77 amino acid forms of IL-8 produced by cultured human cells and cell lines stimulated with IL-1 (PMID: 1593135). The neutralization activity of this antibody towards IL-8 was studied in a rabbit model of severe acute pancreatitis. It was reported that this antibody can reduce the acute lung injury, presumably through inhibition of circulating IL-8 and TNF-alpha, and CD11b/CD18 in lung tissue (PMID: 10189850). This antibody can neutralize rabbit IL-8 in vivo (PMID: 8347559). The effect of neutralizing antibody on IL-8 enhanced infectious cytomegalovirus (CMV) production was examined and it was reported that this antibody reduced virus production nearly to unstimulated levels at 1,000 ng/mL (PMID: 7933146). This antibody was used in the immunohistochemical staining and immuno-localization of IL-8 in tissue sections taken from intestinal loops of patients infected with either M90T or BS176 strains of Shigella flexneri (PMID: 10024597).
