  • Purpose

    Anti-JAM-A [6F4], Rabbit IgG, kappa

    Specificity

This antibody specifically recognizes human junctional adhesion molecule A and does not cross-react with other members of the JAM family. JAM-A is an adherens and tight junction protein expressed by endothelial and epithelial cells. It is recently identified as an antigenic protein (Ag) overexpressed in tumor cells and associated with cancer progression.

    Characteristics

    Original Species of Ab: Mouse

    Original Format of Ab: IgG1

    Purification

    Protein A affinity purified

    Immunogen

The original antibody was generated by immunizing BALB/c mice with MCF-7 cells. The humanized version was generated by grafting CDRs of parental mouse antibody onto human framework regions.

    Isotype

    IgG kappa
  • Application Notes

This antibody was used for the characterization of human junctional adhesion molecule-A (JAM-A) and study of its expression on various cancerous tissues. The binding specificity of this antibody for human JAM-A expressed on CHO cells was confirmed using flow cytometry. JAM-A expression level on cell surface was determined by immunofluorescence staining and visualized by confocal microscopy. This antibody can also be used in the determination of JAM-A protein using western blot. Immunohistochemical analysis using this antibody revealed that JAM-A is overexpressed on breast, lung and kidney tumor tissues. Injections of anti-JAM-A antibody resulted in a significant tumor growth inhibition of xenograft human tumors in vivo. Treatment with monoclonal antibody induced a decrease of the Ki67 expression and downregulated JAM-A levels. This antigen may play a role in interfering with tumor proliferation (PMID: 22886345). This antibody binds the extracellular domain of JAM-A in an ELISA (Goetsch et al, 2011).

    Comment

This chimeric rabbit antibody was made using the variable domain sequences of the original Mouse IgG1 format for improved compatibility with existing reagents assays and techniques.
