    Purpose

    In Vivo Grade Recombinant Anti-mouse CD370 / PD-L1 Bispecific Antibody, Mouse IgG2c LALAPG Kappa (Clone 10B4 / 10F.9G2.1)

    Characteristics

    The rat anti-mouse CD370 (Clec9A or DNGR-1) monoclonal antibody 10B4 (rat IgG2a kappa) reacts with the mouse CD370 protein. CLEC9A (C-type lectin domain family 9 member A), a transmembrane glycoprotein with a single extracellular C-type lectin domain (CTLD), plays a role in the immune response. As an activation receptor, CLEC9A is expressed on myeloid lineage cells, helps process material from dead cells, recognizes filamentous actin when cell membranes are damaged, and helps present antigens to cytotoxic T cells. CLEC9A has an intracellular cytoplasmic tail with potential signaling motifs. CLEC9A is expressed on a subset of BDCA3+ dendritic cells and CD16- monocytes in humans, and is expressed on CD8+ conventional DCs and plasmacytoid DCs in mice. The rat anti-mouse PD-L1 monoclonal antibody 10F.9G2 (rat IgG2b kappa) reacts with the mouse PD-L1 protein (programmed death ligand-1, B7-H1 or CD274), a member of the B7 family of the Ig superfamily. PD-1 has two ligands, PD-L1 and PD-L2. It has been shown that in mouse models of melanoma, tumor growth can be transiently arrested via treatment with the anti-mouse PD-1 and anti-mouse PD-L1 antibodies which block the interaction between the PD-L1 protein and its receptor PD-1 protein. The 10F.9G2 monoclonal antibody blocks the binding of the mouse PD-L1 protein to the mouse PD-1 protein. It is under investigation how the recombinant anti-mouse CD370 / PD-L1 bispecific antibodies (10B4 / 10F.9G2) behave different from the individual monoclonal antibodies and their combination. Our recombinant anti-mouse CD370 / PD-L1 bispecific antibodies have a part (variable regions) or complete amino acid sequences of the rat anti-mouse CD370 antibody (hybridoma clone name or number: 10B4) and the rat anti-mouse PD-L1 antibody (hybridoma clone name or number: 10F.9G2).
