    Protocol

mouse CD48 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from rat specific for CD48 has been precoated onto 96-well plates. Standards(NSO, F23-S217) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for CD48 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the mouse CD48 amount of sample captured in plate.

    Assay Procedure

Aliquot 0.1 mL per well of the 4000pg/mL, 2000pg/mL,1000pg/mL, 500pg/mL, 250pg/mL, 125pg/mL, 62.5pg/mL mouse CD48 standard solutions into the precoated 96-well plate. Add 0.1 mL of the sample diluent buffer into the control well (Zero well). Add 0.1 mL of each properly diluted sample of mouse cell culture supernates, serum or plasma(heparin, EDTA) to each empty well. See "Sample Dilution Guideline" above for details. It is recommended that each mouse CD48 standard solution and each sample be measured in duplicate.

    Background

Protein Function: Ligand for CD2. Might facilitate interaction between activated lymphocytes. Probably involved in regulating T-cell activation.

Background: CD48 antigen(Cluster of Differentiation 48), also known as BLAST-1 or SLAMF2, is a protein that in humans is encoded by the CD48 gene. CD48 is a member of the CD2 subfamily of the immunoglobulin superfamily(IgSF) which includes SLAM(signaling lymphocyte activation molecules) proteins, such as CD84, CD150, CD229 and CD244. It is mapped to 1q23.3. CD48 is found on the surface of lymphocytes and other immune cells, dendritic cells and endothelial cells, and participates in activation and differentiation pathways in these cells. The encoded protein does not have a transmembrane domain, however, but is held at the cell surface by a GPI anchor via a C-terminal domain which maybe cleaved to yield a soluble form of the receptor.

    Synonyms: CD48 antigen,BCM1 surface antigen,BLAST-1,HM48-1,MRC OX-45 surface antigen,SLAM family member 2,SLAMF2,Signaling lymphocytic activation molecule 2,sgp-60,CD48,Cd48,Bcm-1,

    Full Gene Name: CD48 antigen

Cellular Localisation: Cell membrane, Lipid-anchor, GPI-anchor.
