    Before using Kit, spin tubes and bring down all components to bottom of tube. Duplicate well assay was recommended for both standard and sample testing.

    Comment

    Tissue Specificity: Expressed at low level in specific organs of the adult animal such as spleen, heart, lung and brain. During gastrulation, expressed in the forming mesoderm. In later stages of the developing heart, expression is restricted to the truncus arteriosus.

    Protocol

    mouse Cripto ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from rat specific for Cripto has been precoated onto 96-well plates. Standards (NSO, R26-Q150) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Cripto is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the mouse Cripto amount of sample captured in plate.

    Assay Procedure

    Aliquot 0.1 mL per well of the 4000pg/mL, 2000pg/mL, 1000pg/mL, 500pg/mL, 250pg/mL, 125pg/mL, 62.5pg/mL mouse Cripto standard solutions into the precoated 96-well plate. Add 0.1 mL of the sample diluent buffer into the control well (Zero well). Add 0.1 mL of each properly diluted sample of mouse cell culture supernates, serum or plasma(heparin, EDTA) to each empty well. See "Sample Dilution Guideline" above for details. We recommend that each mouse Cripto standard solution and each sample is measured in duplicate.

    Background

    Protein Function: Could play a role in the determination of the epiblastic cells that subsequently give rise to the mesoderm.

    Background: Teratocarcinoma-derived growth factor 1 is a protein that in humans is encoded by the TDGF1 gene. It is mapped to 3p23-p21. The protein is an extracellular, membrane-bound signaling protein that plays an essential role in embryonic developmentand tumor growth. Mutations in this gene are associated with forebrain defects. Pseudogenes of this gene are found on chromosomes 2, 3, 6, 8, 19 and X. Alternate splicing results in multiple transcript variants.
