    Before using Kit, spin tubes and bring down all components to bottom of tube. Duplicate well assay was recommended for both standard and sample testing.

    Tissue Specificity: Highly expressed by dendritic cells in mesenteric and peripheral lymph nodes. Significant expression in spleen (T cell zone or periarteriolar lymphatic sheath) and Peyer patches. Low expression in thymus.

    Protocol

    mouse CCL19 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from rat specific for CCL19 has been precoated onto 96-well plates. Standards(Expression system for standard: E.coli, Immunogen sequence: G26-S108) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for CCL19 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the mouse CCL19 amount of sample captured in plate.

    Assay Procedure

    Aliquot 0.1 mL per well of the 1000pg/mL, 500pg/mL, 250pg/mL, 125pg/mL, 62.5pg/mL, 31.3pg/mL, 15.6pg/mL mouse CCL19 standard solutions into the pre-coated 96-well plate. Add 0.1 mL of the sample diluent buffer into the control well (Zero well). Add 0.1 mL of each properly diluted sample of mouse cell culture supernates, serum or plasma(heparin, EDTA) to each empty well. See "Sample Dilution Guideline" above for details. We recommend that each mouse CCL19 standard solution and each sample is measured in duplicate.

    Protein Function: Strongly chemotactic for naive (L-selectinhi) CD4 T- cells and for CD8 T-cells and weakly attractive for resting B- cells and memory (L-selectinlo) CD4 T-cells. May play a role in promoting encounters between recirculating T-cells and dendritic cells and in the migration of activated B-cells into the T-zone of secondary lymphoid tissues. Binds to chemokine receptor CCR7. Binds to atypical chemokine receptor ACKR4 and mediates the recruitment of beta-arrestin (ARRB1/2) to ACKR4.

    Background: Chemokine(C-C motif) ligand 19 (CCL19) is a small cytokine belonging to the CC chemokine family that is also known as EBI1 ligand chemokine (ELC) and macrophage inflammatory protein-3-beta (MIP-3-beta). Using PCR of human-rodent hybrids, the CCL19 gene was mapped to human chromosome 9p13, and the CCL19 gene is not part of the CC chemokine gene cluster on chromosome 17. The cytokine encoded by this gene may play a role in normal lymphocyte recirculation and homing. It also plays an important role in trafficking of T cells in thymus, and in T cell and B cell migration to secondary lymphoid organs.
