Purpose
- Purified Protein in ready-to-use SDS sample buffer.
-
Characteristics
- AnxA1-selective antibodies were generated against a peptide taken from the human protein, N-terminal region corresponding to AA 20-38. The AnxA1-selective antibodies are affinity purified on an immobilized antigen based affinity matrix, the isolated antibodies were then stabilized in antibody stabilization buffer for long-term storage. The anti- AnxA1-selective antibodies are fully characterized for applications in western blotting and ELISA at the recommended dilutions. The supplier provides AnxA1 Western blot positive control samples in SDS-PAGE sample buffer.

Application Notes
-
Antibodies were tested in ELISA and Western blotting applications at 1:500 dilution using ABIN1686556 samples. Antibody dilutions for these antibodies are for reference only, investigators are expected to determine the optimal conditions. Application of this antibody in other protocols has not yet tested.

IMM & IP pull-down assays: n.d.
IHC: n.d.
Investigators using this antibody in protocols other than listed above can request a complimentary sample of this antibody. n.d. not necessarily means the antibody is not suitable for that application, it simply means we have not yet characterized the antibody for that application. -

Background
- Annexins are a structurally related family of calcium /phospholipid-binding and actin regulatory proteins from evolutionary conserved multigene family. The name is so called since their main property is to bind i.e. to annex. Annexins bind to anionic phospholipids in calcium (Ca2+) dependent manner and share a conserved domain of 4 repeat sequences of 70 residues long containing several Ca2+ binding sites. They are involved in the regulation of a wide range of molecular and cellular processes such as providing a membrane scaffold, trafficking, and organization of vesicles, fibrinolysis, coagulation, inflammation and apoptosis. Dysregulation of annexin family members is reported in numerous cancers affects cellular behaviors such as proliferation, invasiveness and cancer-related signaling pathways suggesting that Annexins play important roles in tumoral development & progression. Annexin A1 (AnxA1, lipocortin-1) is a glucocorticoid-regulated 42- kDa protein. AnxA1, an endogen mediator of the anti-inflammatory effects of glucocorticoids, is involved in a broad range of molecular and cellular processes, including acute and chronic inflammation, cell growth and differentiation, cell proliferation and migration. AnxA1 promotes metastasis and actin reorganization, facilitating an EMT-like (Epithelial-Mesenchymal Transition) switch, thus allowing efficient cell migration and invasion of metastatic breast cancer cells. AnxA1 replicates anti-inflammatory effects of glucocorticoids and is involved in the modulation of T-cell function and the adaptive immune responses relevant to rheumatoid arthritis.
