Purification tag / Conjugate
- This GPR124 protein is labelled with DYKDDDDK Tag,Strep Tag.
-
Purpose
- Human AGRA2-Strep full length protein-synthetic nanodisc
-
Characteristics
- Unlike other membrane scaffold protein (MSP) Nanodisc on the market, our synthetic Nanodisc can be prepared directly from the cells. The polymers used during this process have a dual function. It dissolves the cell membranes, like the detergent, and uses cellular phospholipids to form Nanodisc around the membrane proteins. The target protein embedded Nanodiscs can then be purified.

Advantages:
- Highly purified membrane proteins
- High solubility in aqueous solutions
- High stability
- Proteins are in a native membrane environment and remain biologically active
- No detergent and can be used for cell-based assays
- No MSP backbone proteins
- Intolerant to acids and high concentrations of divalent metal ions
-
Restrictions
- For Research Use only
-
-
-
Format
- Lyophilized
-
Reconstitution
- Please see Certificate of Analysis for specific instructions of reconstitution
-
Buffer
- Lyophilized from nanodisc solubilization buffer (20 mM Tris-HCl, 150 mM NaCl, pH 8.0). Normally 5 % - 8 % trehalose is added as protectants before lyophilization.
-
Storage
- -20 °C,-80 °C
-
Storage Comment
- Store at -20°C to -80°C for 12 months in lyophilized form. After reconstitution, if not intended for use within a month, aliquot and store at -80°C (Avoid repeated freezing and thawing). Lyophilized proteins are shipped at ambient temperature.

Background
-
GPR124, TEM5
Endothelial receptor which functions together with RECK to enable brain endothelial cells to selectively respond to Wnt7 signals (WNT7A or WNT7B) (PubMed:28289266, PubMed:30026314). Plays a key role in Wnt7-specific responses, such as endothelial cell sprouting and migration in the forebrain and neural tube, and establishment of the blood-brain barrier (By similarity). Acts as a Wnt7-specific coactivator of canonical Wnt signaling: required to deliver RECK-bound Wnt7 to frizzled by assembling a higher-order RECK-ADGRA2-Fzd-LRP5-LRP6 complex (PubMed:30026314). ADGRA2-tethering function does not rely on its G-protein coupled receptor (GPCR) structure but instead on its combined capacity to interact with RECK extracellularly and recruit the Dishevelled scaffolding protein intracellularly (PubMed:30026314). Binds to the glycosaminoglycans heparin, heparin sulfate, chondroitin sulfate and dermatan sulfate (PubMed:16982628).[UniProtKB/Swiss-Prot Function] -
