Description ErbB3 is a member of the EGFR/erbB receptor tyrosine kinase family. ErbB3 lacks intrinsickinase activity due to an amino acid substitution within the ATP binding site. Uponheterodimerization with the other erbB family members, erbB3 becomes phosphorylated atmultiple tyrosine residues that represent docking sites for downstream signaling proteins,e. g. PI3K, grb7 and shc. The erbB3 cytoplasmic domain contains 6 docking sites for PI3K. Thus, erbB3 is a potent activator of the PI3K/PKB signaling pathway.
Specificity This antibody recognizes erbB3 phosphorylation status independent at 185 kDa in Westernblot. Species: Human. Others not tested. Add. Information: Includes Positive Control: Cell lysate from untreated SW620 cells (see protocols).

Application Details

Protocol Positive Control: Cell lysate from untreated SW620 cellsFormulation: Lyophilized cell lysate from serum starved SW 620 cellsReconstitution: Reconstitute by addition of 200 µl H2O. After complete solubilization add200 µl 2x SDS-PAGE sample buffer, mix and incubate at 90 °C for 5 min. Aliquote and store frozen. Avoid repeated freeze/thaw cycles.
Application Notes Western Blot: 0.5 ug/ml for HRPO/ECL detection. Recommended blocking buffer: Casein/Tween 20. Positive Control: Cell lysate from untreaed SW620 cells. (See Protocols)Immunohistochemistry on Paraffin Sections. Immunocytochemistry: (1-10 ug/ml). Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
Buffer 1 ml PBS / 0.09% Sopdium Azide / PEG and Sucrose.Reconstitution: Restore with 1 ml H2O (15 min, RT).
Storage Store lyophilized (preferably in a desiccator) at -20°C and reconstituted (aliquote andfreeze in liquid nitrogen) at -80°C. Avoid repeated freezing and thawing. Thaw aliquots at 37°C. Thawed aliquots may be stored at 4°C up to 3 months. Shelf life: one year from despatch.
