Specificity
- AE-1 detects in immunoblots distinct bands of 40 kDa, 45 kDa, 46 kDa, 48 kDa, 50 kDa, (50' kDa), 51 kDa, 55 kDa, and 56.5 kDa with all Ip's lower than 5.7. These MW's are believed to correspond with keratin 19, 18, 17, 16, 15, 14, 13, 12 and 10, respectively. Species Reactivity: Human, bovine, chicken, mouse, rabbit, and rat. Others not tested.

Application Notes
- Immunohistochemistry: (Formalin-fixed / Paraffin-embedded sections) Use at 1:150-1:200 for 30 min. at 37 °C. Higher dilutions can be used if sections are incubated at 4 °C overnight. Western blot: Use at 1:100. The optimal dilution for a specific application should be determined by the researcher.

Background
- This antibody recognizes most of the acidic (type I) keratins. Each epithelium contains at least one acidic keratin. AE-1 is broadly reactive with almost all epithelial and their neoplasms. The antibody exhibits heterogenous staining within a given epithelium, the detailed staining pattern varies depending on the differentiated and/or growth state of the individual cells. This antibody together with MC-976 (clone AE-3) has been used for studying the immuno-localization of epidermal keratins, tissue distribution and epithelial development. AE-1 and AE-3 have also been used extensively, individually or mixed, for distinguishing carcinoma from non-carcinoma.
