  • Application Notes
    IHC, ICC (frozen or formalin–fixed paraffin-embedded (FFPE) tissue sections, cell smears) 1:50-1:200 using streptavidin-Biotin system or polymer system. Prediluted antibody is used directly without dilution. FFPE tissue sections require antigen retriever (boiling tissue in 10 mM citrate pH 6.0 for 10-20 minutes, followed by cooling at RT for 15-20 minutes). The optimum dilution should be determined by the individual lab. Positive Control: Human tonsil, liver. Cellular Localization: Cytoplasmic staining of macrophages.

    Background
    Alpha-1-Antitrypsin (AAT) is a 51 kDa glycoprotein synthesized in the liver. It is a serum protease inhibitor in neutrophil elastase, collagenase and chymotrypsin. This protein is a serum trypsin inhibitor, the correct name is alpha-1-proteinase inhibitor (A1PI). Disorders of this enzyme include, deficiency, in a hereditary disorder, this causes degradation of lung tissue and causes pulmonary emphysema, it also causes chronic liver disease and hepatocarcinoma. AAT is present in normal liver cells, histocytes, monocytes and a variety of epithelial and mesenchymal tumors. Increase in serum levels of AAT is also reported in rheumatoid arthritis, bacterial infection, vasculistis and carcinomatosis. Intended USE: Immunohistochemistry (IHC) and Immunocytchemistry (ICC)
