    Specificity
    The antiserum does not cross-react with any other component of human plasma. Inter-species cross-reactivity is a normal feature of antibodies to plasma proteins since they frequently share antigenic determinants. of this antiserum has not been tested in detail.

    IgG fraction of polyclonal rabbit antiserum to human lysozyme
    Purification
    Adsorption: Immunoaffinity adsorbed using insolubilized antigens as required, to eliminate antibodies cross-reacting with other with other plasma proteins. The use of insolubilized adsorption antigens prevents the presence of excess adsorbent protein or immune complexes in the antiserum. The IgG (7S) fraction is isolated and purified from the antiserum and contains the bulk of the defined antibody specificity. It is free of other serum proteins as tested by immunoelectrophoresis and double radial immunodiffusion.
    Immunogen
    Lysozyme, a bacteriolytic enzyme, has a molecular weight of about 15,000. Its concentration in plasma is low, in milk the concentration is reported to be between 5 and 220 mg/ml. Also tears contain a considerable amount of lysozyme. Highly purified lysozyme is isolated from pooled milk. Freund’s complete adjuvant is used in the first step of the immunization procedure.

  • Application Notes
    The cytochemical grade allows the use in different types of highly sensitive immunoassays on appropriately treated cell and tissue substrates, in radioimmunoassay, for the production of immuno-conjugates with a selected marker, to prepare immunoaffinity adsorbents by coupling to an artificial carrier, in non-isotopic methodology based on solid phase immunochemistry (e.g. ELISA), both as catching antibody and detection reagent, in Western blotting. This product is not pre-diluted. The optimum working dilution of each product should be established by titration before being used. Working dilutions for histochemical and cytochemical use are usually between 1:100 and 1:250, in ELISA and comparable non-precipitating antibody-binding assays are between 1:500 and 1:5,000.

    Background
    The reactivity of the antiserum is restricted to lysozyme. In immunoelectrophoresis and radial immuno-diffusion (Ouchterlony), using various antiserum concentrations against normal human milk and tears a single precipitin line is obtained which shows a reaction of identity with the precipitin line obtained with purified lysozyme. No reaction is obtained with any other plasma protein component or serum
