Description
|
ErbB3 is a member of the EGFR/erbB receptor tyrosine kinase family. ErbB3 lacks intrinsickinase activity due to an amino acid substitution within the ATP binding site. Uponheterodimerization with the other erbB family members, erbB3 becomes phosphorylated atmultiple tyrosine residues that represent docking sites for downstream signaling proteins,e. g. PI3K, grb7 and shc. The erbB3 cytoplasmic domain contains 6 docking sites for PI3K. Thus, erbB3 is a potent activator of the PI3K/PKB signaling pathway.

Cell lysate from untreated SW620Description: Cell lysate from untreated SW620 cells, colorectal adenocarcinoma (Human)Format: Lyophilized cell lysate from starved cells SW 620 cells. Reconstitution: Restore by addition of 200 µl H20. After complete solubilization add 200 µl2x SDS-PAGE sample buffer, mix and incubate at 90°C for 5 min. Storage: Aliquote and store frozen. Avoid repeated freeze/thaw cycles.

Immunoblotting: 0.5 µg/ml for HRPO/ECL detection. Recommended blocking buffer: Casein/Tween 20 based blocking and blot incubationbuffer AS00002BU-N or AS00002BU-L. Included Positive Control: Cell lysate from untreated SW620 cells (See Protocols for moredetails). Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
