This antibody has been used on Chlamydomonas cells expressing endogenous Fox1 and on HEK293 cells transiently expressing Fox1. Cells were resuspended in PBS plated on poly-lysine coated coverslips, fixed in 3.7% formaldehyde. Blocked in 3% BSA in PBS. 1:500 dilution of anti-FOX1 in 3% BSA. Secondary antibody was anti-rabbit Alexa fluor 488 1:300.

- Background
- A copper-containing protein in Chlamydomonas reinhardtii is a plasma membrane ferroxidase, encoded by the FOX1 gene, involved in Fe assimilation. FOX1 is most highly related to the mammalian homologues hephaestin and ceruloplasmin, its occurrence and pattern of expression in Chlamydomonas indicate a role for copper in iron assimilation in a photosynthetic species.
