ELISA: In a sandwich ELISA (assuming 100 μL/well), a concentration of 2-4 μg/mL of thisantibody will detect at least 100 pg/mL of recombinant Human MDC when used withbiotinylated antigen affinity purified anti-Human MDC antibody (cat. PP1049B) as thedetection antibody at a concentration of at least 1 μg/mL. Western Blot: To detect Human MDC by Western Blot analysis this antibody can be used ata concentration of 0.25-0.50 μg/mL. Used in conjunction with compatible secondaryreagents the detection limit for recombinant hMDC is 0.25-0.50 ng/lane, undernon-reducing conditions. Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity ofhMIG (100 ng/mL), a concentration of 5.0-10.0 μg/mL of this antibody is required. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.

Store the antibody prior to reconstitution at -20 °C. Following reconstitution the antibody can be stored at 2-8 °C for one month or at -20 °C for longer.
The MDC gene is one of several Cys-Cys (CC) cytokine genes which are clustered on the q arm of chromosome 16. Cytokines are a family of secreted proteins involved in immunoregulatory and inflammatory processes. The CC cytokines are proteins characterized by two adjacent cysteines. The cytokine encoded by MDC displays chemotactic activity for natural killer cells, chronically activated T lymphocytes, monocytes and dendritic cells. It has no chemoattractant activity for eosinophils, neutrophils and resting T lymphocytes and also displays a mild activity for primary activated T lymphocytes. The product of this gene binds to chemokine receptor CCR4.
