  • Application Notes

    Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity ofHuman IL-9 (5.0 ng/mL), a concentration of 1.0-2.0 μg/mL of this antibody is required. Sandwich ELISA: To detect Human IL-9 by Sandwich ELISA (using 100 μL/well antibodysolution) a concentration of at least 0.5-2.0 μg/mL of this antibody is required. This antigenaffinity purified antibody, in conjunction with Biotinylated anti-Human IL9 antibody(PP1031B1 or PP1031B2) as a Detection antibody, allows the detection of at least 0.2-0.4 ng/well of recombinant Human IL-9. Western Blot: To detect hIL-9 by Western Blot analysis this antibody can be used at aconcentration of 0.1-0.2 μg/mL. Used in conjunction with compatible secondary reagentsthe detection limit for recombinant hIL-9 is 1.5-3.0 ng/lane, under either reducing ornon-reducing conditions.
    Other applications not tested.
    Optimal dilutions are dependent on conditions and should be determined by the user.

    Background

    Interleukin 9 (IL9) is a cytokine preferentially produced by T helper type 2 lymphocytes and active on various cell types such as T and B lymphocytes, mast cells and haemopoietic progenitors, but no effect of this cytokine has been reported so far on mononuclear phagocytes. IL9 stimulates cell proliferation and prevents apoptosis. The IL9 receptor (IL 9R) belongs to the haemopoietic receptor superfamily and its signal transduction involves mainly the (JAK/STAT) pathway. Some studies have implicated IL9 as an important cytokine in a number of Th2 cytokine mediated pathologies, in particular asthma.Synonyms: IL-9, P40 cytokine, T-cell growth factor P40
