    ELISA: (Direct): To detect mEGF by direct ELISA (using 100 μL/well antibody solution) aconcentration of 0.25-1.0 μg/mL of this antibody is required. This biotinylated polyclonalantibody, in conjunction with compatible secondary reagents, allows the detection of atleast 0.2-0.4 ng/well of recombinant mEGF. (Sandwich): To detect mEGF by sandwich ELISA (using 100 μL/well antibody solution) aconcentration of 0.25-1.0 μg/mL of this antibody is required. This biotinylated polyclonalantibody, in conjunction with Polyclonal Anti-Murine EGF (PP1126P) as a capture antibody,allows the detection of at least 0.2-0.4 ng/well of recombinant mEGF. Western Blot: To detect mEGF by Western Blot analysis this antibody can be used at aconcentration of 0.1-0.2 μg/mL. Used in conjunction with compatible secondary reagentsthe detection limit for recombinant mEGF is 1.5-3.0 ng/lane, under either reducing ornon-reducing conditions.
    Other applications not tested.
    Optimal dilutions are dependent on conditions and should be determined by the user.

    Background

    Epidermal growth factor (EGF) has a profound effect on the differentiation of specific cells in vivo and is a potent mitogenic factor for a variety of cultured cells. The EGF precursor is believed to exist as a membrane bound molecule which is proteolytically cleaved to generate the 53 amino acid peptide hormone that stimulates cells to divide. EGF exerts its actions by binding to the EGFR, a 170 kDa protein. Epidermal growth factor (EGF) is a key growth factor regulating cell survival. Through its binding to cell surface receptors, EGF activates an extensive network of signal transduction pathways that include activation of the PI3K/AKT, RAS/ERK and JAK/STAT pathways.
