  • Specificity

    Assay by immunoelectrophoresis resulted in a single precipitin arc against purified and partially purified Carbonic Anhydrase I [Human Erythrocytes]. Cross reactivity against Carbonic Anhydrase I from other tissues and species may occur but have not been specifically determined.

    This product was prepared from monospecific antiserum by a delipidation and defibrination.

  • Application Notes

    Suitable for immunoblotting (western or dot blot), ELISA, immunoprecipitation and mostimmunological methods requiring high titer and specificity. This product has been assayed against 1.0 μg of Carbonic Anhydrase I [HumanErythrocytes] in a standard sandwich ELISA using Peroxidase conjugated Affinity Purifiedanti-Goat IgG [H&L] (Rabbit) and (ABTS (2,2'-azino-bis-[3-ethylbenthiazoline-6-sulfonicacid]) as a substrate for 30 minutes at room temperature. A working dilution of 1: 1,000 to1: 5,000 is suggested for this product.
    Other applications not tested.
    Optimal dilutions are dependent on conditions and should be determined by the user.

    Store vial at 4° C prior to restoration. Restore with deionized water (or equivalent), centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4 °C as an undiluted liquid. For extended storage add glycerol to 50% and then aliquot contents and freeze at -20° C or below. Avoid cycles of freezing and thawing.

    Background

    Carbonic Anhydrase I (CAI) is a single polypeptide chain metalloenzyme of molecular weight 29 kDa. It is present mainly in erythrocytes at a concentration of about 12 mg per gram haemoglobin. Its main enzyme function is to catalyse the reversible hydration of carbon dioxide. It also catalyses the hydrolysis of certain esters. The twelve active CA isozymes are thought to regulate a variety of cellular functions including several processes in the reproductive systems. A mutational deficiency has been described but patients exhibit no adverse chemical symptoms.
