    Sequence

    S-S-G-K-G-Y-G-pS-Q-V-A-Q-Q-F-E Remarks: The RFA complex consists of a heterotrim er of RFA1, RFA2 and RFA3, 69, 36 and 13 kda chains respective ly. Yeast RFA2 is phosphoryl ated at the G1/S transition and is dephosphor ylated at mitosis.

    Specificity

    This affinity purified antibody is directed against yeast RFA2. The product was affinity purified from monospecific antiserum by immunoaffinity purification. Antiserum was first purified against the phosphorylated form of the immunizing peptide. The resultant affinity purified antibody was then cross-adsorbed against the non-phosphorylated form of the immunizing peptide. This phospho specific polyclonal antibody is specific for phosphorylated pS122 of yeast RFA2. Reactivity with non-phosphorylated yeast RFA2 is minimal by ELISA.

    Immunogen

    This affinity purified antibody was prepared from whole rabbit serum produced by repeated immunizations with a synthetic peptide corresponding to aa 115-129 (of 273) of yeast DNA Replication Factor A (RFA2).
  • Application Notes

    This affinity-purified antibody was tested for use in western blotting against RFA2containing cell lysates and by ELISA against the immunizing peptide. Reactivity in otherimmunoassays is unknown. This product was assayed by western blotting against RFA2containing cell lysates. A 1: 500 to 1: 1000 dilution is recommended for this assay. Thisproduct was also assayed by ELISA against 0.1 μg of the phosphorylated form of theimmunizing peptide. A 1: 8000 to 1: 20000 dilution of the antibody is recommended for thisassay. Equivalent signal is seen against the non-phosphorylated form of the peptide whenthe antibody is diluted to 1: 300.
    Other applications not tested.
    Optimal dilutions are dependent on conditions and should be determined by the user.
