    ELISA: Direct: To detect hsCD40L by direct ELISA (using 100 μL/well antibody solution) aconcentration of 0.25 - 1.0 μg/mL is required. In conjunction with compatible secondaryreagents, it allows the detection of at least 0.2 - 0.4 ng/well of recombinant hsCD40L. Sandwich: To detect hsCD40L by sandwich ELISA (using 100 μL/well antibody solution) aconcentration of 0.25 - 1.0 μg/mL is required. In conjunction with Polyclonal Anti-HumansCD40L as a capture antibody, it allows the detection of at least 0.2 - 0.4 ng/well ofrecombinant hsCD40L. Western Blot: To detect hsCD40L by Western Blot analysis this antibody can be used at aconcentration of 0.1 - 0.2 μg/mL. Used in conjunction with compatible secondary reagentsthe detection limit for recombinant hsCD40L is 1.5 - 3.0 ng/lane, under either reducing ornon-reducing conditions.
    Other applications not tested.
    Optimal dilutions are dependent on conditions and should be determined by the user.

    Background

    CD40 ligand (CD40L) is a 33 kDa type II membrane glycoprotein expressed mainly on the cell surface of activated T lymphocytes, but also exists as a soluble form extracellularly. CD40L is the ligand for CD40, a member of the TNF superfamily, which is expressed on the cell surface of B cells, macrophages/monocytes, dendritic cells, vascular endothelial cells, and epithelial cells. CD40L plays an important role in B cell proliferation, antibody class switching, modulation of apoptosis in the germinal center through interaction with B cells expressing CD40, and activation of CD4+ T cells.
