Western Blotting (WB), Immunohistochemistry (IHC), Immunohistochemistry (Paraffin-embedded Sections) (IHC (p)), Immunoprecipitation (IP)
  • Specificity

Reacts with all six known vertebrate isoforms of actin (MW ~42kD). It also reacts with two cytoplasmic actins (beta, gamma) which are highly homologous to one another, but which differ from the muscle actins by about 25 amino acid residues. This antibody is highly recommended for monitoring total protein load on Western blots. Species cross-reactivity: Human, bovine, mouse, canine, porcine, rabbit, rat, chicken, Dictyostelium discoideum, Physarum polycephalum.

    Purification

    Protein G purified

    Immunogen

    Chicken gizzard actin.

    Type of Immunogen: Purified protein

    Isotype

    IgG1 kappa
  • Application Notes

    Approved: IHC, IHC-P (0.5 μg/mL), IP, WB (0.5 μg/mL)

Usage: Suitable for use in Immunoprecipitation, Immunohistochemistry and Western Blot. Immunoprecipitation (Denatured verified): 2 μg/mg protein lysate. Use Protein G. Western Blot: 0.5 μg/mL for 2hrs at room temperature. Immunohistochemistry (formalin/paraffin): 1:400 for 30 min. Staining of formalin-fixed, paraffin-embedded tissues requires boiling tissue sections in 10 mM citrate buffer,  pH 6.0, for 10-20 min. followed by cooling at RT for 20 min. Positive control: HeLa cells, skeletal muscle. The applications listed have been tested for the unmodified form of this product. Other forms have not been tested.
