Immunoprecipitation (IP)
  • Specificity

The antiserum does not cross-react with any other human plasma proteins as tested in gel-diffusion techniques. Inter-species cross-reactivity is a normal feature of antibodies to plasma proteins, since homologous proteins of different species frequently share antigenic determinants. of this antiserum has not been tested in detail, however in double radial immunodiffusion a weak reaction with mouse has been observed.

    Characteristics

Precipitating polyclonal goat antiserum to human properdin factor B

    Purification

Adsorption: Immunoaffinity adsorbed using insolubilized antigens as required, to eliminate antibodies reacting with other human serum proteins. The use of insolubilized adsorption antigens prevents the presence of excess adsorbent protein or immune complexes in the antiserum.

    Immunogen

Properdin factor B is a heat-labile serum protein active in the alternative complement activation pathway. It is binding to the C3b component of complement. The protein has a molecular weight of 92,000 and its mean concentration in human plasma is 200 µg/ml. It has been isolated as a homogenous protein for use in antiserum production. Freund’s complete adjuvant is used in the first step of the immunization procedure.

    Isotype

    IgG
  • Application Notes

In precipitating techniques for the identification and quantitative determination of properdin factor B in body fluids. The presence of non-precipitating antibodies has not been assayed. This does not exclude the use of the antiserum in more sensitive techniques if proper controls are included.

    Restrictions

    For Research Use only
  • Format

    Lyophilized

    Concentration

Total protein and IgG concentration in the antiserum are comparable to those of pooled normal goat serum. No foreign proteins added. Antibody titre: Preciptin titre not less than 1:8 when tested against normal human serum in agar block immunodiffusion tit

    Buffer

Delipidated, heat inactivated, lyophilized, stable whole serum.

    Preservative

    Without preservative

    Storage

    4 °C/-20 °C

    Storage Comment

The lyophilized antiserum is shipped at ambient temperature and may be stored at +4°C, prolonged storage at or below -20°C. Reconstitute the lyophilized antiserum by add ing 1 ml sterile distilled water. Dilutions may be prepared by adding phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the antiserum. Diluted antiserum should be stored at +4°C, not refrozen, and preferably used the same day.
  • Target

    Complement Factor B (CFB)

    Alternative Name

    Properdin Factor B

    Background

Tested in immunoelectrophoresis against human EDTA plasma a single precipitin line is obtained in the beta-2 region. The antiserum does not react with any other protein component of human serum or plasma

    Pathways

Complement System, Proton Transport, Ribonucleoside Biosynthetic Process
