Application Details

Application Notes: Uncoupled sdAb for doing maleimide conjugations. You can chose the position of the ectopic cysteine (N-terminal, C-terminal or both ends).
Western Blotting not recommended. The sdAbs tend to recognize native protein conformation only.

Protocol: Maleimide Labelling Protocol

• Remove TCEP and exchange buffer to 10 mM KPi, 300 mM NaCl, 2 mM EDTA, pH 6.0 by size exclusion chromatography
• Before labelling, add 1/10 vol 1M Tris/HCl pH 8.0
• On ice, add 1.5- to 5- fold molar excess of fluorophore as maleimide
• Quickly vortex
• Overlay with Argon
• Place on ice for 90 min
• Separate excess of dye from the labelled sdAb e.g. by size exclusion chromatography

Reconstitution: Unconjugated sdAbs are lyophilized from 10 mM KPi pH 6.0,150 mM NaCl, 1 mM EDTA. Reconstitute with 250 µL pure water. Use unconjugated sdAbs immediately after reconstitution.
