Neutralization of human IL-31 bioactivity. It is recommended to establish the optimal antibody concentration for the assay system used. Neutralization Neutralization assay: Neutralization of human IL-31 induced IL-6 production (see graph). To determine the neutralizing capacity and ND50, mixtures of human recombinant IL-31 (50 ng/ ml) and various concentrations of mAb MT313 were added to human aortic endothelial cells that had been pre-stimulated with recombinant human IFN-γ (10 ng/mL). Following overnight incubation, cell supernatants were quantified in IL-6 ELISA. Neutralization dose 50 (ND50): MT313 neutralized the effect of human IL-31 at a ND50value of 0.1-0.6 μg/mL when 50 ng/mL of human IL-31 was present. ND50 is defined as the concentration of antibody that results in 50 % inhibition of the biological response under the experimental conditions specified.
