    Anti-XCR1 [5G7], Mouse IgG2b, kappa

    Specificity

    This antibody binds human XCR1, more particularly it strongly binds amino acids 'PESTTFFYYDLQ' of the human XCR1. XRC1 is a receptor for chemokines SCYC1 and SCYC2 and expressed on 80 % of CD8+ the dendritic cells. It also acts as a receptor for XCL1/Lymphotactin. It transduces a signal by increasing the intracellular calcium ions level.

  • Application Notes

    This antibody is capable of binding human XCR1-EGFP-expressing B300.19 cells in a flow cytometry analysis. The determination of the binding epitope of this antibody was done using ELISA (US9371389). This antibody exhibits a significant effect in treating rheumatoid arthritis in the experiment of delayed-type hypersensitivity reaction induced by Mycobacterium butyricum delayed-type hypersensitivity reaction. The neutralizing activity of this antibody against human lymphotactin-induced migration of human XCR1-expressing cells was evaluated by in vitro chemotaxis assay. It was reported that this antibody completely inhibited cell migration at a concentration of about 3 μg/mL (US9371389). The effect of this antibody on ear swelling of DNFB (Dinitrofluorobenzene)-sensitized mice was done and it was reported that this antibody caused significant suppression of ear swelling 24 after induction. This antibody was also reported to show showed a very low reactivity to human CX3CR1-EGFP-expressing B300.19 cells (US9371389). The original antibody with saporin-conjugated secondary antibody showed growth suppression of human XCR1-EGFP-expressing B300.19 cells. The humanized version of this antibody is also available on request (US9371389).
