Background

Nploc4, or nuclear protein localization protein four homolog, is a highly conserved protein involved in protein degradation and cellular stress response pathways. It acts as a cofactor in the disassembly of polyubiquitinated protein complexes, particularly in cooperation with the ATPase p97 also known as VCP. Nploc4 plays an essential role in extracting ubiquitinated substrates from membranes and protein complexes to facilitate their degradation by the proteasome.
In zebrafish, Nploc4 is expressed in multiple tissues and is especially active in proliferative or metabolically active cells. The zebrafish Nploc4 protein is an ortholog of the human NPLOC4 gene. Both versions of the protein share strong sequence and functional conservation, indicating that zebrafish can serve as a reliable model for studying NPLOC4 related biology.
Functionally, Nploc4 is part of a complex with Ufd1 and p97, and this complex is critical for endoplasmic reticulum associated degradation, chromatin remodeling, and cell cycle regulation. Disruption of Nploc4 function in zebrafish leads to defects in protein homeostasis and developmental abnormalities, especially in systems that are sensitive to proteotoxic stress such as the nervous system and musculature.
Given its central role in maintaining cellular protein quality control and its conservation with human NPLOC4, zebrafish Nploc4 is commonly used in developmental studies and disease models related to protein aggregation, neurodegeneration, and cellular stress adaptation.
