    Description:

    Endophilin-1 is a member of the Endophilin family, a group of proteins characterized by the presence of BAR (Bin/Amphiphysin/Rvs) and SH3 (Src Homology 3) domains.

    The Endophilin family is primarily divided into two subfamilies. Endophilin A , that includes subunits A1, A2 and A3, is predominantly involved in membrane remodeling processes such clathrin-mediated and clathrin-independent endocytic pathways, especially in neurons. The Endophilin B subfamily includes subunits B1 and B2, and is more implicated in mitochondrial dynamics, autophagy and apoptosis.1,2

    Endophilin-A1 is expressed almost exclusively in the brain, particularly in synaptic terminals in neurons, consistent with a neuronal specialization for membrane trafficking and synaptic function. It is essential for synaptic vesicle endocytosis, where it recruits and organizes proteins at sites of high membrane curvature to facilitate the formation and recycling of clathrin-coated vesicles at synaptic terminals. It is critical for dendritic spine morphogenesis, synaptic stabilization, and plasticity, supporting both excitatory and inhibitory synaptic organization.3,4

    Changes in Endophilin-1 expression are linked to several neurodegenerative disease, most notably, Alzheimer's (AD) and Parkinson disease. Increased expression of Endophilin-1 has been observed in the brains of patients with AD and in transgenic mouse models of the disease.5,6

    Endophilin-1 was identified as a Parkinson's disease risk locus and in addition, it was found to be upregulated in a mouse model of the disease.7,8

    In summary, Endophilin-A1 not only serves as a compelling target for understanding synaptic biology but also holds promise as a biomarker and therapeutic candidate in neurodegenerative and neuropsychiatric diseases.
