    Protocol

    human Furin ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Furin has been precoated onto 96-well plates. Standards(NSO, D108-E715) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Furin is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human Furin amount of sample captured in plate.

    Assay Procedure

    Aliquot 0.1 mL per well of the 20000pg/mL, 10000pg/mL, 5000pg/mL, 2500pg/mL, 1250pg/mL, 625pg/mL, 312pg/mL human Furin standard solutions into the precoated 96-well plate. Add 0.1 mL of the sample diluent buffer into the control well (Zero well). Add 0.1 mL of each properly diluted sample of human cell culture supernates, tissue homogenates or cell lysates to each empty well. See "Sample Dilution Guideline" above for details. It is recommended that each human Furin standard solution and each sample be measured in duplicate.

    Protein Function: Furin is likely to represent the ubiquitous endoprotease activity within constitutive secretory pathways and capable of cleavage at the RX(K/R)R consensus motif. .

    Background: Furin(Fur), also known as PACE, is a protein that in humans is encoded by the FURIN gene. The protein encoded by this gene is an enzyme which belongs to the subtilisin-like proprotein convertase family. The FUR gene is located approximately 1 kb upstream of the FES gene on chromosome 15q25-q26. This gene is thought to play a role in tumor progression. Furin is one of the proteases responsible for the proteolytic cleavage of HIV envelope polyprotein precursor gp160 to gp120 and gp41 prior to viral assembly. Furin is enriched in the Golgi apparatus, where it functions to cleave other proteins into their mature/active forms. Expression of furin in T-cells is required for maintenance of peripheral immune tolerance

Shuttles between the trans-Golgi network and the cell surface. Propeptide cleavage is a prerequisite for exit of furin molecules out of the endoplasmic reticulum (ER). A second cleavage within the propeptide occurs in the trans Golgi network (TGN), followed by the release of the propeptide and the activation of furin.
