    The Alpha2-macroglobulin (rat)-ELISA is designed for the quantitative determination of Alpha2-macroglobulin (A2M) in rat body fluids and cell culture supernatants.

  • Plate
    Pre-coated
    Protocol
    A2M has been pre-coated onto a microplate. During incubation the binding of an anti-A2M(rat)-antibody to the wells is inhibited by A2M from samples/standards. The immobilized antibody is detected using a HRP-labelled secondary antibody. After washing away any unbound antibody, a substrate solution is added to the wells and color develops in proportion to the amount of antibody conjugate. The absorption at 450 nm is reverse proportional to the A2M concentration.
    Assay Procedure

    Incubation of samples/standards with antibody conjugate: 50 µL + 50 µL/well 60 min, room temperature with shaking
    Wash Incubation of detection antibody (HRP-labeled): 100 µL/well 60 min with shaking
    Wash Substrate incubation: 100 µL/well 20 min, room temperature
    Add stop solution: 100 µL/well Read at 450 nm
