Protocol: Useful in Sandwich ELISA for Quantitative Detection of Antigen. Aliquot 0.1 mL per well of the 50 ng/mL, 25 ng/mL, 12.5 ng/mL, 6.25 ng/mL, 3.12 ng/mL, 1.56 ng/mL, 0.78 ng/mL human Ficolin-2 standard solutions into the precoated 96-well plate. Add 0.1 mL of the sample diluent buffer into the control well (Zero well). Add 0.1 mL of each properly diluted sample of human cell culture supernates, serum or plasma(heparin) to each empty well. It is recommended that each human Ficolin-2 standard solution and each sample be measured in duplicate.

Background:

Background: Ficolin-2, which was initially identified as L-ficolin, is a protein that in humans is encoded by the FCN2 gene. The product of this gene belongs to the ficolin family of proteins. This family is characterized by the presence of a leader peptide, a short N-terminal segment, followed by a collagen-like region, and a C-terminal fibrinogen-like domain. It is mapped to 9q34.3. This gene is predominantly expressed in the liver, and has been shown to have carbohydrate binding and opsonic activities. Serum concentration of ficolin-2 varies considerably in healthy individuals. FCN2 promoter polymorphisms were associated with marked changes in theficolin-2 serum concentration.
