Protocol
- ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. An antibody has been precoated onto 96-well plates. Standards and test samples are added to the wells, a biotinylated detection antibody specific for target is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the target amount in sample captured in plate.

Background: Meprin A subunit alpha also known as endopeptidase-2 or PABA peptide hydrolase is the alpha subunit of the meprin A enzyme that in humans is encoded by the MEP1A gene. Meprin A subunit alpha, encoded by the MEP1A gene, is a component of the meprin A metalloproteinase complex, which is involved in extracellular matrix remodeling and proteolysis. Meprin A is a zinc-dependent endopeptidase found predominantly in the kidneys, intestines, and other tissues. It plays a crucial role in various physiological processes, including inflammation, tissue repair, and the processing of bioactive peptides. The alpha subunit, specifically, is a part of the hetero-oligomeric enzyme complex, which combines with the beta subunit to form an active enzyme. Abnormalities in meprin A function have been associated with several pathological conditions, including kidney diseases and cancer.
