Protocol
- ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. An antibody has been precoated onto 96-well plates. Standards and test samples are added to the wells, a biotinylated detection antibody specific for target is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the target amount in sample captured in plate.

Background: Angiopoietin-related protein 7 is a protein that in humans is encoded by the ANGPTL7 gene. This gene is mapped to 4, 4 E2. Angiopoietin-like Protein 7 (ANGPTL7), also known as cornea-derived transcript 6 (CDT6), is a secreted 45 kDa protein that contains a coiled-coil domain (aa 30-110) and a fibrinogen-like domain (aa 113-334). ANGPTL7 is expressed in the stromal layer of the cornea, and it forms disulfide-linked homotetramers. It contributes to corneal morphogenesis by suppressing angiogenesis and promoting the deposition of extracellular matrix components. ANGPTL7 is up-regulated in glaucoma and accumulates in the aqueous humor. Mature mouse ANGPTL7 shares 88 % and 98 % sequence identity with human and rat ANGPTL7.
