Purpose
- Nuclease ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of nucleases, including SuperNuclease® and Benzonase®, in cell culture supernatants and purified biological products while nuclease is supplied to remove host cell DNA during manufacturing.

Specificity
- This ELISA kit has been validated with SuperNuclease®, Benzonase® and RayBio® Universal Nuclease, and should react with other Serratia marcescens nuclease products but not tested. Both non-denatured and denatured nuclease can be detected by this kit.

Protocol
-
- Prepare all reagents, samples and standards as instructed in the manual.
- Add 100 μL standard or sample to each well. Incubate 2.5 hours at room temperature.
- Add 100 μL prepared biotin antibody to each well. Incubate 1 hour at room temperature.
- Add 100 μL prepared Streptavidin solution to each well. Incubate 45 minutes at room temperature.
- Add 100 μL TMB One-Step Substrate Reagent to each well. Incubate 30 minutes at room temperature.
- Add 50 μL Stop Solution to each well. Read at 450 nm immediately
