Application Notes
- This protein carries a human IgG1 Fc tag at the C-terminus, followed by an Avi tag (Avitag™). The protein has a calculated MW of 54.9 kDa. The protein migrates as 70-80 kDa under reducing (R) condition due to glycosylation.

Ready-to-use Avitag™ biotinylated protein:
The product is exclusively produced using the Avitag™ technology. Briefly, a unique 15 amino acid peptide, the Avi tag, is introduced into the recombinant protein during expression vector construction. The single lysine residue in the Avi tag is enzymatically biotinylated by the E. Coli biotin ligase BirA.
This single-point enzymatic labeling technique brings many advantages for commonly used binding assays. The biotinylation happens on the lysine residue of Avi tag, and therefore does NOT interfere with the target protein's natural binding activities. In addition, when immobilized on an avidin-coated surface, the protein orientation is uniform because the position of the Avi tag in the protein is precisely controlled. -

Background

Synonyms: CD33,SIGLEC3,gp67,
Description: Myeloid cell surface antigen CD33 is also known as SIGLEC3, Siglecs (sialic acid binding Ig-like lectins) and GP67, is a single-pass type I membrane protein which belongs to the immunoglobulin superfamily and SIGLEC (sialic acid binding Ig-like lectin) family. Human CD33 / Siglec-3 cDNA encodes a 364 amino acid (aa) polypeptide with a hydrophobic signal peptide, an N--terminal Ig--like V--type domain, one Ig--like C2--type domains, a transmembrane region and a cytoplasmic tail. CD33 / Siglec-3 usually considered myeloid-specific, but it can also be found on some lymphoid cells. In the immune response, CD33 / Siglec-3 may act as an inhibitory receptor upon ligand induced tyrosine phosphorylation by recruiting cytoplasmic phosphatase(s) via their SH2 domain(s) that block signal transduction through dephosphorylation of signaling molecules. CD33 / Siglec-3 induces apoptosis in acute myeloid leukemia. -
