Biological Activity Comment
- NGF is purified in three forms: the 7S, 2.5S and β. The 7S 130 kDa form occurs naturally in mouse submaxillary glands and is a multimeric protein composed of two α, one β and two γ subunits. The biologically active subunit is the β, which is a 26 kDa dimer composed of two identical chains held together by hydrophobic interactions. The α chain stabilizes the 7S complex and binds two zinc ions per 7S complex as a cofactor at the β-γ interface. The γ chain is an arginine specific protease, it may also have plasminongen activator activity as well as mitogenic activity for chick embryo fibroblasts1,2.

Reconstitution
- Centrifuge the vial at 10,000 x g for 5 minutes before opening to ensure all material is collected at the bottom. The lyophilized product may be difficult to visualize. Add solvent directly to the centrifuged vial and dissolve by gently tapping, tilting, or rolling the vial. Avoid vigorous vortexing, a brief, light vortex of 1-2 seconds is acceptable if necessary. Reconstitute in sterile, high-purity water at a concentration of at least 0.1 mg/mL. For long-term storage, aliquot the solution into small, single-use vials and store at -20 °C. Before use, thaw the relevant vial(s) and dilute to the desired working concentration in your working buffer. Centrifuge all product preparations before use. It is recommended to prepare fresh working solutions just before use. Avoid multiple freeze-thaw cycles to maintain optimal biological activity.
