Protein Size: Native humanBMP-6 isgenerated by the proteolytic removal of the signal peptide and propeptide, the molecule has a calculated molecular mass of approximately17 kDa. Recombinant BMP-6 is a disulfide-linked homodimeric protein consisting of two 140 amino acid residue subunits,and migrates as an approximately34 kDa protein under non-reducing conditions and as a 17 kDa protein under reducing conditions in SDS-PAGE.

Sequence Position: Optimized DNA sequence encoding Human Bone Morphogenetic protein-6 mature chain was expressed in Escherichia Coli.

Sterility: 0.2 μm filtered

Endotoxin Level: Endotoxin content was assayed using a LAL gel clot method. Endotoxin level was found to be less than 0.1 ng/μg(1EU/μg).

Biological Activity Comment: The ED(50) was determined by its ability to induce alkaline phosphatase production by mouse ATDC5 chondrogenic and was found to be in the range of0.1-0.5 ug/ml.

Reconstitution: A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.

Buffer: Recombinant Bone morphogenetic Protein-6was lyophilized from a 0.2 μm filtered solution in 2.5 % glycine, 0.5 % sucrose, 0.01 % Tween80, 5 mM Glutamic acid, pH 4.5.

Storage: 4 °C,-20 °C

Storage Comment: The lyophilized protein is stable for at least 2 years from date of receipt at -20°C. Upon reconstitution, this cytokine can be stored in working aliquots at 2°- 8°C for one month, or at -20°C for six months, with a carrier protein without detectable loss of activity. Avoid repeated freeze/thaw cycles.
