Protein Background: DDX54 (ATP-dependent RNA helicase DDX54) is a nucleolar DEAD-box RNA helicase that functions as a key regulator of transcription and ribosome biogenesis. It is recruited to gene promoters, including those of nuclear receptors and stress-responsive genes, where it acts as a transcriptional co-regulator to modulate their expression. Furthermore, DDX54 plays a direct role in the nucleolar stages of ribosomal RNA (rRNA) processing and maturation, which is essential for the proper assembly of ribosomes. Its activity is also implicated in the cellular stress response, where it contributes to the reorganization of nucleolar architecture and halts rRNA synthesis to conserve energy. Dysregulation of DDX54 has been associated with cancers, underscoring its importance in fundamental cell growth and proliferation pathways.
