Horse (Equine) Cytochrome P450, Family 3, Subfamily A, Polypeptide 4 (CYP3A4) interaction partners

  1. It was concluded that CYP3A related metabolism in horse is not solely dependent on the expression of the enzyme but also on adequate levels of NADPH P450 reductase (show POR Proteins) and cytochrome b(5 (show CYB5A Proteins)).

Human Cytochrome P450, Family 3, Subfamily A, Polypeptide 4 (CYP3A4) interaction partners

  1. Metabolism of amiodarone to desethylamiodarone by CYP1A1 (show CYP1A1 Proteins) or CYP3A4 plays an important role in the hepatocellular toxicity of amiodarone.

  2. Report roles for CYP3A4 and CYP2C9 (show CYP2C9 Proteins) in sequential two-step bioactivation of diclofenac to reactive p-benzoquinone imines.

  3. SLCO1B1 (show SLCO1B1 Proteins) rs113681054, SLCO1B1 (show SLCO1B1 Proteins)*5 (rs4149056), CYP3A4*1G (rs2242480), and CYP3A4*5 (rs776746) polymorphisms had no effect on the PK/PD of ticagrelor in healthy Chinese volunteers

  4. Data suggest that CYP3A4 (cytochrome P-450 3A4) is involved in proliferation, migration, invasion, and colony formation abilities of hepatocellular carcinoma cells; in HEPG2 cells, the naturally occurring flavonoid rutin (an antioxidant anticarcinogen) inhibits progression (inhibits proliferation, migration, etc.), promotes apoptosis, and down-regulates expression of CYP3A4.

  5. effector-dependent conformational equilibrium between open and closed states of CYP3A4

  6. The combination of an in vivo drug-drug interaction (DDI) study with NVS123 and the potent CYP3A4 inhibitor ritonavir in hCYP3A4 mice along with physiologically based pharmacokinetic modeling modeling allowed for a prospective prediction of the extent of DDI and boost effect in humans, which was subsequently confirmed in a clinical study.

  7. Suggest that benzbromarone is a mechanism-based CYP3A4 inhibitor.

  8. This study found significant changes in the disposition of lidocaine hydrochloride in native healthy Tibetan and Han Chinese subjects living at a high altitude in comparison to healthy Han Chinese subjects living at low altitude, which might be due to significant decreases in the activity and protein and mRNA expression of CYP3A4 under chronic high altitude hypoxia condition.

  9. CYP2D6 and CYP3A activities were shown to have moderate contribution on clearance of tramadol enantiomers.

  10. decreases of CES and CYP3A4 expression and enzymatic activities induced by Fluoxetine are through decreasing PXR (show NR1I2 Proteins) and increasing DEC1 (show BHLHE40 Proteins) in HepG2 cells

CYP3A4 Protein Profile

Protein Summary

This gene encodes a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases that catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This protein localizes to the endoplasmic reticulum and its expression is induced by glucocorticoids and some pharmacological agents. This enzyme is involved in the metabolism of approximately half the drugs in use today, including acetaminophen, codeine, cyclosporin A, diazepam and erythromycin. The enzyme also metabolizes some steroids and carcinogens. This gene is part of a cluster of cytochrome P450 genes on chromosome 7q21.1. Previously another CYP3A gene, CYP3A3, was thought to exist\; however, it is now thought that this sequence represents a transcript variant of CYP3A4. Alternatively spliced transcript variants encoding different isoforms have been identified.
