  • Application Notes

    Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity of RatIL-1 beta (0.20 ng/mL), a concentration of 0.09-0.14 μg/mL of this antibody is required. Sandwich ELISA: To detect Rat IL-1 beta by sandwich ELISA (using 100 μL/well antibodysolution) a concentration of 0.5-2.0 μg/mL of this antibody is required. This antigen affinitypurified antibody, in conjunction with biotinylated Anti-Rat IL-1 beta (PP034B1 or PP034B2)as a detection antibody, allows the detection of at least 0.2-0.4 ng/well of recombinant RatIL-1 beta. Western blot: To detect Rat IL-1 beta by Western blot analysis this antibody can be used ata concentration of 0.1-0.2 μg/mL. Used in conjunction with compatible secondary reagentsthe detection limit for recombinant Rat IL-1 beta is 1.5-3.0 ng/lane, under either reducing ornon-reducing conditions.
    Other applications not tested.
    Optimal dilutions are dependent on conditions and should be determined by the user.

    Background

    IL1-beta is a proinflammatory cytokine produced in a variety of cells including monocytes, tissue macrophages, keratinocytes and other epithelial cells. Both IL1A and IL1B binds to the same receptor and has similar if not identical biological properties. These cytokines have a broad range of activities including, stimulation of thymocyte proliferation, by inducing IL-2 release, B-cell maturation and proliferation, mitogenic FGF-like activity and the ability to stimulate the release of prostaglandin and collagenase from synovial cells. However, whereas IL-1B is a secreted cytokine, IL-1A is predominantly a cell-associated cytokine.Synonyms: Catabolin, IL-1 beta, IL1 beta, IL1B, IL1F2
