    NeutralizationTo yield one-half maximal inhibition [ND50] of the biological activity of Rat TNF-alpha(2.0 ng/mL), a concentration of 0.07 - 0.1 μg/mL of this antibody is required. ELISAIndirect: To detect Rat TNF-alpha by indirect ELISA (using 100 μL/well antibody solution) aconcentration of 0.5 - 2.0 μg/mL of this antibody is required. This antibody, in conjunctionwith compatible secondary reagents, allows the detection of at least 0.2 - 0.4 ng/well ofrecombinant Rat TNF-alpha. Sandwich: To detect Rat TNF-alpha by sandwich ELISA (using 100 μL/well antibodysolution) a concentration of 0.5 - 2.0 μg/mL of this antibody is required. This antibody, inconjunction with biotinylated Anti-Rat TNF-alpha (PP039B1 or PP039B2) as a detectionantibody, allows the detection of at least 0.2 - 0.4 ng/well of recombinant Rat TNF-alpha. Western BlotTo detect Rat TNF-alpha by Western Blot analysis this antibody can be used at aconcentration of 0.1 - 0.2 μg/mL. Used in conjunction with compatible secondary reagentsthe detection limit for recombinant Rat TNF-alpha is 1.5 - 3.0 ng/lane, under either reducingor non-reducing conditions.
    Other applications not tested.
    Optimal dilutions are dependent on conditions and should be determined by the user.

    Background

    TNF-alpha is a pleiotrophic pro-inflammatory cytokine secreted by various cells including adipocytes, activated monocytes, macrophages, B cells, T cells and fibroblasts. It belongs to the TNF family of ligands and signals through two receptors, TNFR1 and TNFR2. TNF-alpha is cytotoxic to a wide variety of tumor cells and is an essential factor in mediating the immune response against bacterial infections.
