  • Application Notes
    Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity ofHuman sIL-2RA (10.0 μg/mL), a concentration of 1.0-1.5 μg/mL of this antibody is required,in the presence of 0.5 ng/mL recombinant hIL-2. ELISA: In a Sandwich ELISA (assuming 100μL/well), a concentration of 4.0-8.0 μg/mL of thisantibody will detect at least 100 pg/mL of recombinant Human sIL-2RA when used inconjunction with compatible secondary reagents. Western Blot: To detect Human sIL-2RA by Western Blot analysis this antibody can be usedat a concentration of 0.20-0.40 μg/mL. Used in conjunction with compatible secondaryreagents the detection limit for recombinant Human sIL-2RA is 2.0-4.0 ng/lane, underreducing or non-reducing conditions.
    Other applications not tested.
    Optimal dilutions are dependent on conditions and should be determined by the user.

    Background
    The Interleukin 2 Receptor alpha and beta chains, together with the common gamma chain, constitute the high affinity IL2 receptor present on activated T and B cells, thymocyte subset, pre B cells and T regulatory cells. Homodimeric alpha chains result in low affinity receptor, while homodimeric beta chains produce a medium affinity receptor. Normally an integral membrane protein, soluble IL2 Receptor alpha has been isolated and determined to result from extracellular proteolysis.
