Specificity
This antibody reacts with an internal epitope of MVP/LRP (p110) accordingly to an increase in the number of vault particles. The MVP/LRP (p110) is strongly overexpressed in various human non-P-glycoprotein MDR tumor cell lines.
Western blotting: Use 1:50 dilution (chemi-luminescence). Immunohistochemistry: Use 1:20-50 dilution on 4% paraformaldehyde fixed cytospin preparations or frozen tissue sections. The following pretreatment is recommended: 10 min 20mM glycine (pH 7.5) and 10 min 6N guanidine hydrochloride in 50 mM Tris-HCl (pH 7.5). MVP-37 performs well on paraffin-embedded tissue sections.
