Specificity
This antibody reacts with an internal epitope of LRP/MVP (p110) accordingly to an increase in the number of vault particles. LRP is strongly overexpressed in various human non-P-glycoprotein MDR tumor cell lines.
Western blotting: Use at a 1:50 dilution (chemi-luminescence). Immunohistochemistry: Use at a 1:20 to 1:50 dilution on 4% paraformaldehyde fixed cytospin preparations or frozen tissue sections. The following pretreatment is recommended: 10 min. in 20 mM glycine, pH 7.5, and 10 min. in 6 N guanidine hydrochloride in 50 mM Tris-HCl, pH 7.5. MVP-37 performs well on paraffin-embedded tissue sections. The optimal dilution for a specific application should be determined by the researcher.
