Application Notes: Immunohistochemistry (IHC) 1:50-1:200 using polymer system or streptavidin-biotin system. Formalin-fixed paraffin-embedded (FFPE) tissue sections and cell smears do require antigen retrieval (boiling tissue in 10 mM citrate buffer, pH 6.0 for 10 minutes, followed by cooling to RT for 10-15 minutes) The optimum dilution should be determined by the individual lab Positive Control: Human muscle or sarcomas. Background: Desmin is a type III intermediate filament found near the Z line in sarcomere. Desmin is only expressed in vertebrates, however homologous proteins are found in many organisms. It is a 52kD protein that is a subunit of intermediate filaments in skeletal muscle tissue, smooth muscle tissue, and cardiac muscle tissue. Antibodies to desmin are useful in identification of tumors of myogenic origin.
