  • Application Notes

    Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity ofmIL-12 (0.3 ng/mL), a concentration of 0.012 - 0.2 μg/mL of this antibody is required. ELISA: To detect mIL-12 by direct ELISA (using 100 l/well antibody solution) aconcentration of at least 0.5 μg/mL of this antibody is required. This antigen affinitypurified antibody, in conjunction with compatible secondary reagents, allows the detectionof 0.2-0.4 ng/well of recombinant mIL-12. Western Blot: To detect mIL-12 alpha by Western Blot analysis this antibody can be used ata concentration of 0.1- 0.2 μg/mL. Used in conjunction with compatible secondary reagentsthe detection limit for recombinant mIL-12 is 1.5-3.0 ng/lane, under either reducing ornon-reducing conditions. Immunohistochemistry: We recommend use of 1 g per ml as a starting dilution. Paraffinsections may require pretreatment like Proteinase K or Trypsin predigestion to improvestaining.
    Other applications not tested.
    Optimal dilutions are dependent on conditions and should be determined by the user.

    Background

    Interleukin 12 (IL12) can act as a growth factor for activated T and NK cells, enhance the lytic activity of NK/lymphokine activated killer cells, and stimulate the production of IFN gamma by resting PBMC. IL12 induces IFN gamma, TNF alpha production in T and NK cells, costimulation of PBL proliferation, proliferation/differentiation of TH1 T lymphocytes. It is downregulated by IL10, homodimeric p40 antagonistic to functional p70 heterodimer.
