Neutralization Activity: This antibody has been reported to be useful for the neutralization of recombinant human IL-10. Neutralization activity may be measured with a proliferation assay using 5 ng/mL recombinant human IL-10 (Cat. No. 554611), co-stimulated with 50 pg/mL recombinant mouse IL-4 (Cat. No. 550067), to stimulate MC/9 or D36 cells as indicator cells (i.e preincubation of the antibody with recombinant human IL-10 for 60 min)

- Application Notes
- ELISA: The purified format of the JES3-19F1 antibody (Cat. No. 554705) has been reported to be useful as a capture antibody in sandwich ELISA for human IL-10. Purified JES3-19F1 antibody can be paired with the biotinylated JES3-12G8 antibody (Cat. No. 554499) and with recombinant human IL-10 (Cat. No. 554611) as the standard. Purified JES3-19F1 antibody should be titrated to determine the optimal concentration for ELISA capture. An appropriate range of concentrations for the human IL-10 standard for obtaining a linear standard curve is 15 pg/mL to 2000 pg/mL. For testing human IL-10 in complex biological fluids such as serum or plasma, the BD OptEIA™ Human IL-10 ELISA Set (Cat. No. 555157) or Kit (Cat. No. 550613) is recommended.

- Background
- The JES3-19F1 antibody reacts with human (IL-10). The immunogen used to generate the JES3-19F1 hybridoma was recombinant human IL-10 expressed in COS cells. This is a neutralizing antibody. This antibody also reacts with viral IL-10.
